Builder is more relaxed and focused on exploring and building. Less zombies, more loot, and you can hit more than one zombie at a time. Survivor is classic survival mode. Standard zombies, normal loot, and can only hit one zombie at a time. Apocalypse, I haven't played as much of, but it's harder survival mode. Using all the usual suspects for negative traits: ThinSkinned, High Thirst, Slow Healer, Underweight, Hearty Appetite, Injured, Pacifist, Prone to Illness, Smoker, Weak Stomach, Cowardly. Postive traits I use are : Cat's Eyes, Dextrous, Fast Reader (I know, I can skip time but man I just love powering through books), Wakeful, Antique Collector ... The best traits in Project Zomboid are Short Sighted, High Thirst and Weak Stomach, they are examples of traits that cause little to no inconvenience and or are easy to fix. Short Sighted only affects foraging radius and makes no difference to actual perception sight.
